초록

이 연구는 PLS 시행 후 일부 농약성분이 토양에 잔류하고 후작물로 흡수이행되어 일률기준 0.01 mg/kg 을 초과할 우려가 있어 후작물 잔류시험의 필요성을 확인하고자 수행되었다. 농약안전사용기준에 따라 입제를 처리한 후 녹색꽃양배추와 양상추, 상추를 재배하여 수확기 토양 및 작물 중 농약잔류량을 조사하였다. 그리고 입제 처리 후 31일에 상추를 정식하여 흡수이행된 잔류량을 조사하였다. 이 시험기간동안 토양은 지속적으로 점적관수하였다. 녹색꽃양배추 중 cadusafos 및 양상추 중 pencycuron 잔류량은 정량한계 0.01 mg/kg 미만이었으며, 녹색꽃양배추 중 fluopyram과 양상추 중 flutolanil은 기준량과 배량처리구에서 0.01 mg/kg을 초과하였다. 토양 중 잔류량은 0.2-3.58mg/kg으로 이론적 처리농도의 40% 이상 이었다. Hexaconazole 입제 처리 2일 후 정식한 상추의 잔류량은 <0.01mg/kg이었지만, 입제 처리 후 점적관수를 하고 31일차에 정식한 상추에는 0.02 mg/kg이 잔류하였다. 이 시험을 통해 토양에 처리된 후 천천히 농약이 방출되는 입제의 경우, 당해작물에는 잔류하지 않더라도 토양에 상당량이 잔류할 수도 있어 토양수분이 충분한 조건에서는 후작물로 이행될 가능성이 있음을 확인할 수 있었다. 따라서 후작물에 잔류될 가능성이 높은 농약들은 후작물 잔류시험을 통하여 후작물 식재일 및 후작물 잔류허용기준을 설정해야 될 것으로 생각된다.

Abstract

This study was carried out to confirm the necessity of a residual test after PLS because some pesticide components remain in the soil and are absorbed into the succeeding crop, which may exceed 0.01 mg/kg on a default MRL. Broccoli, head lettuce and lettuce were cultivated after applying pesticide granules according to the standards for safe use of pesticides, and pesticide residues in the soil and crops at harvest period were investigated. On the 31st day after granulation treatment, lettuce was planted. The soil was drip irrigation during the test period. The residual amounts of cadusafos in broccoli and pencycuron in head lettuce were less than 0.01 mg/kg of the quantitative limit, and fluopyram in broccoli and flutolanil in head lettuce exceeded 0.01 mg/kg in the reference amount and doubling treatment. The residual amount in the soil was 0.2-3.58 mg/kg, which was more than 40% of the theoretical treatment concentration. When the lettuce was planted on the 2nd day after the hexaconazole granule treatment, the residual amount was <0.01 mg/kg in harvest. But when the plot was drip-irrigated for 1 month after treatment and lettuce was planted on the 31st day, hexaconazole was 0.02 mg/kg in harvest. Through this test, it was confirmed that in the case of granules that were released slowly after treatment in the soil, a significant amount of pesticide could remain in the soil even if the pesticide was not detected in the crop. And if sufficient water is supplied to the soil, it is likely to be transferred to the succeeding crop. Therefore, it is considered that the plant back intervals and the MRL for the rotational crops on pesticides with a high possibility of remaining in rotational crops should be established through the pesticide residue test for succeeding crop.

서 론

토양에 잔류하는 농약은 휘발되거나 토양 입자와의 직접 접촉에 의해 작물의 잎, 열매 등의 지상부로 전이될 수도 있지만, 뿌리를 통한 흡수경로가 훨씬 중요한데 뿌리와의 접촉에 의하여 분배·흡수되거나 토양수분에 용해된 상태로 물과 함께 흡수되어 물관을 통하여 지상부(주로 잎)로 이동한다. 토양에서 증기 또는 용액상태로 존재하는 유기화합물은 식물뿌리를 통해 흡수될 수도 있는데 대부분의 경우는 확산에 의한 수동적 흡수이고(Collins et al., 2006), 수경조건에서는 뿌리 주변 용액 중 농도와 뿌리 내 용액 중 농도의 평형, 친유성인 뿌리의 지질부분으로의 흡수 또는 흡착 정도에 따라 결정된다(Briggs et al., 1983). 이러한 특성을 이용하여 농약의 흡수·이행에 관련한 수리모형을 활용한 예측연구가 꾸준히 진행되어 왔다(Briggs et al., 1982, 1983; Hung and Mackay, 1997; Legind et al., 2011; Shinoda, 2016).

그러나 농약이 토양에서 작물체로 흡수·이행되는 정도는 농약의 분해성, 흡착성, 물에 대한 용해도 등 농약의 특성 뿐만 아니라, 토양의 유기물, 수분, 온도, 미생물 활성 등 재배환경 및 작물의 생리적 특성과 생육상태 등 다양한 요인에 의하여 복합적인 영향을 받는다. 따라서 작물잔류에 대한 예측모형은 현상을 설명하거나 주요 변수가 무엇인지를 결정하는 수단으로는 활용 가능하지만, 농약이 토양으로부터 전이되어 작물에 잔류되는 정도를 결정하는 데는 한계가 있다.

따라서 국내에서는 토양에서 작물로 이행되어 잔류문제가 야기되는 농약에 대한 해결방안을 찾는 과정에서 주로 흡수·이행시험을 활용하고 있다. 인삼에 대해서는 tolclofosmethyl (Noh et al., 2012; Kim et al., 2014), boscalid (Hwang et al., 2011), cypermethrin 및 diethofencarb (Lee et al., 2009)의 이행 정도가 보고되었고, 무에 대해서는 endosulfan (Park et al., 2004; Hwang et al., 2016)과 dinotefuran (Kwak et al., 2017), 당근에 대해서는 endosulfan과 procymidone (Park et al., 2004), 오이에 대해서는 chlorpyrifos (Hwang et al., 2014)의 이행 정도가 보고된 바 있다. 배추에 대해서는 boscalid와 chlorfenapyr (Jeon et al., 2014), azoxystrobin (Kim et al, 2017), endosulfan과 procymidone (Park et al., 2004)의 이행시험이 수행된 바 있고, 상추에 대해서는 procymidone (Hwang et al., 2017a)과 tricyclazole (Hwang et al., 2017b)의 이행 정도에 대하여 보고된 바 있다. 그리고 chlorpyrifos, endosulfan, thoprophos, fthalide의 배추, 상추, 시금치별 이행 양상을 비교한 결과도 있다(Park et al., 2017). 작물 잎에 잔류하는 농약의 꿀벌에 대한 독성을 판단할 목적으로 수행하는 시험의 결과로 토양에 입제로 처리된 clothianidin, dinotefuran, imidacloprid의 토마토 잎으로의 이행농도를 보고한 경우도 있다(Bae et al., 2013).

한편, 농약등록 평가과정에서는 작물에 사용된 농약이 토양에 잔류하여 후작물에 흡수이행될 가능성을 윤작시험의 결과로 판단하는데(OECD, 2018), 유럽연합과 미국은 농약 성분별로 후작물 대사시험과 단계별 후작물 잔류시험을 실시하여 후작물 잔류허용기준을 설정하거나 농약처리 후 작물 식재일(Plant back intervals)을 농약 라벨에 적시하여 관리하고 있다. FAO 매뉴얼(FAO, 2016)에 따라 JMPR에서는 유럽연합과 미국이 노지재배 조건에서 수행한 후작물 잔류시험으로 후작물의 농약잔류 가능성을 평가하고 있지만, 국내에서는 시설재배 면적이 많은 농업여건을 감안하여 노지보다 강우나 광의 영향을 적게 받는 시설재배지에서 잔류시험을 수행하여 후작물로의 이행가능성을 평가하는 것이 필요할 것이다.

국내에서는 Positive List System 도입 후 농약잔류허용 기준이 설정되지 않은 농산물에 대해서는 일률기준으로 0.01 mg/kg을 적용하고 있어 토양으로부터 후작물로 흡수·이행되어 잔류허용기준을 초과할 우려가 있는 농약에 대한 후작물 잔류 가능성에 대한 검토가 시급한 실정이다. 토양에서 분해가 느리고 많은 양이 처리되는 농약성분을 선정하여 시설재배지 토양에 입제농약으로 처리하고 녹색꽃양배추와 양상추, 상추를 재배하여 수확기 토양 및 작물 중 농약잔류량을 조사하여 후작물 잔류시험의 필요성을 확인하고자 하였다.


재료 및 방법
대상농약 및 작물

대상농약은 시중 농약상에서 구입하여 사용하였으며, 농약처리량, 작물 품종, 재식밀도, 재배이력 및 일평균 온·습도 등은 Table 1에 나타내었다.

녹색꽃양배추와 양상추는 강원도 횡성군 둔내면 현천리의 비닐온실에서 농가관행대로 재배하였는데, 농약을 토양표면에 고르게 살포 하고 쇠스랑으로 토양과 혼화한 1-4일 후에 작물을 정식하였다.

상추도 전북 정읍시 신태인읍 백산리 비닐온실에서 농가 관행대로 재배하였는데, 농약을 토양표면에 살포하고 쇠스랑으로 토양과 혼화한 후 점적호스로 관수하였고 정식 전에 15 cm 깊이로 경운하였다. 재배포장의 토양특성은 Table 2에 나타내었다.

시료채취 및 조제

녹색꽃양배추와 양상추 잔류시험 토양시료는 작물 시료 수확일에 표면에서 1 0 c m 깊이까지 채취하였으며, 채취한 토양은 음건하여 2 mm 체로 거른 후 분석에 사용하였다. 녹색꽃양배추와 양상추 시료는 처리별로 12개체, 2 kg 이상 채취하였다. 각 개체별 가식부위를 4등분하여 한 부분씩을 세절한 후 혼합하여 마쇄하였다. 마쇄한 시료는 지퍼백에 밀봉하여 분석 전까지 –20°C 냉동고에 보관하였다.

상추 잔류시험 토양시료는 수확일에 표면에서 1 5 c m 깊이까지 8지점에서 채취한 것을 혼합하고, 음건하여 2mm 체로 거른 후 분석에 사용하였다. 상추는 연속수확하지 않고 포기째 일시에 수확하였는데 처리별로 12개체, 2 kg 이상을 채취하였다. 줄기를 제거한 시료를 드라이아이스와 혼합하여 마쇄한 후 지퍼백에 밀봉하여 분석 전까지 –10°C 냉동고에 보관하였다.

시약

토양 분석용 농약표준용액으로는 Kemidas (Korea, Suwon)의 1,000 mg/L stock solution을 사용하였으며, 작물 분석에는 분석용 등급의 cadusafos (97.6%, Wako), fluopyram (99.1%, Wako), flutolanil (99.7%, Wako), pencycuron (99.7%, Sigma-Aldrich)을 사용하였다. Hexaconazole과 thifluzamide 표준용액은 각각 1,007mg/L와 997 mg/L stock solution (AccuStandard, USA, New Haven)을 사용하였다. 시료 추출과 기기분석에 사용된 acetone, acetonitrile, methanol은 HPLC급을 사용하였고, sodium chloride와 sodium sulfate anhydrous는 GR급을 사용하였다.

농약 표준검량선 작성

토양 중 cadusafos, fluopyram, flutolanil 및 pencycuron분석을 위한 표준검량선은 0.01, 0.05, 0.1, 0.5 mg/L 4수준으로 working solution을 만든 후 시료분석과정을 거친 토양 무처리 시료 추출액을 이용하여 1:1로 혼합하여 matrix matched standard solution을 조제하였다. 시료분석 전 matrix matched 표준용액을 분석하여 표준검량선을 작성하였다.

녹색꽃양배추와 양상추 중 cadusafos, flutolanil 및 pencycuron 분석을 위한 표준검량선은 0.01, 0.05, 0.1, 0.2, 0.5, 1.0 mg/L로 작성하였다. 녹색꽃양배추 중 cadusafos 분석을 위하여 working solution 40 mg/L 0.25 mL를 분취하여 농축, 건고시키고 시료분석과정을 거친 무처리 시료 추출액 5 mL로 재용해하여 matrix matched standard solution 2 mg/L를 조제하였다. 이 용액을 무처리시료 추출액으로 희석하여 검량선 작성용 표준용액을 조제하였다. 양상추 중flutolanil 분석을 위하여 working solution 20 mg/L 0.5 mL 를 분취하여 농축, 건고시키고 시료분석과정을 거친 무처리시료 추출액 5mL로 재용해하여 matrix matched standard solution 2 mg/L를 조제하였다. 이 용액을 무처리 시료 추출액으로 희석하여 검량선 작성용 표준용액을 조제하였다. 양상추 중 pencycuron 분석을 위하여 working solution 20mg/L 0.5 mL를 분취하여 농축, 건고시키고 시료분석과정을 거친 무처리 시료 추출액 10mL로 재용해하여 matrix matched standard solution 1 mg/L를 조제하였다. 이 용액을 무처리 시료 추출액으로 희석하여 검량선 작성용 표준용액을 조제하였다. 녹색꽃양배추 중 fluopyram 분석을 위하여 0.01, 0.02, 0.05, 0.1, 0.2 mg/L 5수준으로 검량선 작성용 표준용액을 조제하였다. 작물시료의 경우에는 분석 전·후 표준용액을 분석하여 협차방정식으로 표준검량선을 작성하였다.

Hexaconazole과 thifluzamide 정량용 표준검량선은 토양분석의 경우 0.005, 0.01, 0.02, 0.05, 0.1, 0.2 mg/L 5-6수준으로 작성하였고, 상추시료 분석을 위해서는 0.005, 0.01, 0.02, 0.05, 0.1, 0.2 mg/L 6수준으로 작성하였다. 각 농도수준별 working solution을 만든 후 1mL를 분취하여 농축, 건고시키고 시료분석과정을 거친 무처리 시료 추출액 1mL로 재용해하여 matrix matched standard solution을 조제하였다. 시료 분석 전·후 표준용액을 분석하여 협차방정식으로 표준검량선을 작성하였다.

녹색꽃양배추와 양상추시험 토양시료 분석용으로는 1,000 mg/L stock solution을 acetonitrile로 희석하여 10mg/L의 혼합 working solution을 조제하였고, 작물시료 분석용으로는 cadusafos 등 4종 농약을 acetonitrile로 용해하여 조제한 100 mg/L stock solution을 acetonitrile로 희석하여 working solution을 만들었다. 상추시험 토양과 작물시료 분석용으로는 stock solution을 acetonitrile로 희석하여 100mg/L의 working solution을 조제하였다.

기기분석으로 얻어진 chromatogram의 peak 면적을 기준으로 표준검량선을 작성하였다.

Cadusafos, fluopyram, flutolanil 및 pencycuron 잔류분석
토양 분석

토양은 QuEChERS를 변형한 방법을 이용하여 분석하였다. 토양 시료 15 g에 증류수 10mL를 첨가하여 30분간 습 윤화 시킨 후 1% acetic acid를 포함한 acetonitrile 15 mL를 혼합하여 QuEChERS 추출기(1600 MiniG, SPEX sample Prep, Metuchen, USA)를 이용하여 1,300 rpm으로 2분간 추출하였다. 여기에 MgSO4 4 g 및 MgCl2 1 g을 첨가한 후 QuEChERS 추출기를 이용해 1,300 rpm으로 30초간 흔들어 주고, 4°C 냉장조건에서 3,500 rpm (2,691 xg)으로 5분 동안 원심분리하였다. 상징액 8mL를 취하여 15 mL EN-General kit (MgSO4 900 mg, primary secondary amine (PSA) 150mg)에 첨가한 후 1분간 흔들어주고 3,500 rpm (2,664 xg)으로 5분간 원심분리하였다. 상징액을 0.2 μm syringe filter로 여과한 다음 여과액 500 μL와 acetonitrile 500 μL를 혼합하여 LC-MS/MS에 5μL씩 주입하여 정량하였다(Table 3). Cadusafos, fluopyram, flutolanil 및 pencycuron의 경우 peak 면적이 최고 검량농도의 면적을 초과한 경우에는 시료를 희석한 후 재분석하였다.

녹색꽃양배추 중 cadusafos 분석

녹색꽃양배추 시료 20g에 acetone 100 mL와 Hyflo Super Ce을 가하고 homogenizer로 10,000 rpm에서 5분간 마쇄·추출하였다. 추출액을 Advantec No.2 여과지로 여과하고, acetone을 감압·유거한 후 포화식염수 50mL와 증류수 150 mL를 첨가하고 dichloromethane 50 mL로 5분간 2회 분배하였다. 유기용매층을 sodium sulfate anhydrous에 통과시켜 탈수하고, 감압·농축한 다음 건고물을 n-hexane: ethyl acetate (95: 5, v/v) 5 mL로 재용해하였다. SPE Florisil 910mg cartridge를 n-hexane 5 mL로 세정한 후, 추출액 2.5mL를 loading하여 흘려버리고, n-hexane: acetone (70: 30, v/v) 5 mL를 1mL/min. 속도로 흘려 cadusafos를 용출시켰다. 용출액을 질소가스로 농축한 다음, 건고물을 acetonitrile 10mL로 재용해하였다. 최종 시료용액을 0.2 μm PTFE filter로 여과하여 LC-MS/MS에 2 μL씩 주입하여 정량하였다(Table 4).

녹색꽃양배추 중 fluopyram 분석

녹색꽃양배추 시료 20 g에 acetone 100 mL와 Hyflo Super Cel®을 가하고 homogenizer로 10,000 rpm에서 5분간 마쇄·추출하였다. 추출액을 Advantec No.2 여과지로 여과하고, acetone을 감압·유거한 후 포화식염수 50mL와 증류수 150mL를 첨가하고 dichloromethane 50 mL로 5분간 2회 분배하였다. 유기용매층을 sodium sulfate anhydrous에 통과시켜 탈수하고, 감압·농축한 다음 건고물을 n-hexane 10 mL로 재용해하였다. SPE Florisil 910 mg cartridge를 n-hexane 5mL로 세정한 후, 추출액 5mL를 loading하고 n-hexane: acetone (90: 10, v/v) 5 mL를 흘려버린 후, n-hexane: acetone (70: 30, v/v) 5 mL를 1 mL/min. 속도로 흘려 fluopyram을 용출시켰다. 용출액을 질소가스로 농축한 다음, 건고물을 n-hexane 10 mL로 재용해하였다. 최종 시료용액을 GC-ECD에 1 μL씩 주입하여 정량하였다(Table 4).

양상추 중 flutolanil과 pencycuron 분석

양상추 시료 10g에 acetonitrile 100 mL와 Hyflo Super Cel®을 가하고 homogenizer로 10,000 rpm에서 5분간 마쇄·추출하였다. 추출액을 Advantec No.2 여과지로 여과하고, acetonitrile을 감압·유거한 후 포화식염수 50mL와 증류수150 mL를 첨가하고 dichloromethane 50 mL로 5분간 2회 분배하였다. 유기용매층을 sodium sulfate anhydrous에 통과시켜 탈수하고, 감압·농축한 다음 건고물을 acetonitrile 10 mL로 재용해하였다. 최종 시료용액을 0.2 μm PTFE filter로 여과하여 LC-MS/MS에 2 μL씩 주입하여 정량하였다(Table 4).

Hexaconazole 및 thifluzamide 잔류분석
토양 분석

토양 시료 20 g을 250mL 삼각플라스크에 취하고 acetone 100 mL을 첨가하여 30분간 진탕·추출하였다. 추출물을 약 5 g의 Celite 545가 깔린 büchner funnel상에서 감압·여과하고, 이때 acetone 약 30mL로 용기와 잔사를 씻어 앞의 여액과 합하였다. 이 여액을 500mL 분액여두에 옮겨 포화식염수 50mL와 증류수 150mL를 첨가하고 ichloromethane 50 mL로 2회 분배하였다. 유기용매층을 50 g의 anhydrous sodium sulfate에 통과시켜 탈수하고, 40°C 수욕상에서 감압·농축한 다음 건고물을 acetonitrile 20 mL로 재용해하였다. 재용해액 1mL를 취하여 25 mg PSA, 150 mg magnesium sulfate, 25 mg C18e를 넣어둔 tube에 옮겨 담은 뒤 vortex mixer에서 1분간 혼화하고 2,500 rpm (560 xg)으로 3분 동안 원심분리하였다. 상징액을 0.22 μm PTFE filter로 여과하여 LC-MS/MS에 5 μL씩 주입하여 정량하였다(Table 5).

상추 분석

상추 시료 10 g을 conical t ube에 취하고 acetonitrile 10mL, MgSO4 4 g, NaCI 1 g, trisodium citrate dihydrate 1 g, sodium hydrogen citrate sesquihydrate 0.5 g을 첨가하여 2,500 rpm으로 30분간 추출한 다음, 4,000 rpm (3,100 xg)으로 10분간 원심분리하였다. 추출 상징액 1mL을 25mg PSA, 150 mg magnesium sulfate, 25 mg C18e를 넣어둔 tube에 옮겨 담은 뒤 vortex mixer로 1분간 혼합하고, 2,500rpm (560 xg)으로 3분 동안 원심분리하였다. 정제 상징액을 0.22 μm PTFE filter로 여과하여 LC-MS/MS에 5 μL씩 주입하여 정량하였다(Table 5).

분석법 검증
토양 중 농약의 회수율 시험

Cadusafos, fluopyram, flutolanil 및 pencycuron은 해당농약 10.0 mg/L 표준용액을 무처리 토양시료 15 g에 0.4 또는 2.0 mL씩 첨가하여 해당농약의 잔류량이 0.05 또는 0.25mg/kg이 되도록 하였다. Hexaconazole은 0.2mg/L 또는 2mg/L 표준용액을 무처리 토양시료 20 g에 1 mL씩 첨가하여 잔류량이 0.01mg/kg 또는 0.1mg/kg이 되도록 하였다. Thifluzamide은 0.2mg/L 또는 4mg/L 표준용액을 무처리 토양시료 20 g에 1mL씩 첨가하여 잔류량이 0.01 mg/kg 또는 0.2mg/kg이 되도록 하였다. 농약으로 처리된 토양시료를 해당 잔류분석 방법을 적용하여 회수율을 산출하였다.

작물 중 농약의 회수율 시험

Cadusafos는 무처리 녹색꽃양배추 20 g에 4mg/L 표준용액 1mL 또는 40 mg/L 표준용액 0.5mL를 첨가하여 잔류량이 0.2 mg/kg 또는 1.0mg/kg이 되도록 하였다. Fluopyram은 무처리 녹색꽃양배추 20 g에 4mg/L 표준용액 0.5mL또는 40 mg/L 표준용액 0.25 mL를 첨가하여 잔류량이 0.1mg/kg 또는 0.5mg/kg이 돠도록 하였다. Flutolanil은 4mg/L 또는 20 mg/L 표준용액을 무처리 양상추 10 g에 0.5mL씩 첨가하여 잔류량이 0.2 mg/kg 또는 1.0mg/kg이 되도록하였다. Pencycuron은 무처리 양상추 10 g에 2mg/L 표준용액 1mL 또는 20 mg/L 표준용액 0.5mL를 첨가하여 잔류량이 0.2mg/kg 또는 1.0mg/kg이 되도록 하였다. Hexaconazole과 thifluzamide는 해당농약 0.1mg/L 또는 1.0mg/L 표준용액을 무처리 상추 10 g에 1mL씩 첨가하여 잔류량이 0.01 mg/kg 또는 0.1 mg/kg이 되도록 하였다. 농약으로 처리된 작물시료를 해당 잔류분석 방법을 적용하여 회수율을 산출하였다. Cadusafos의 1.0mg/kg 수준 회수율은 peak 면적이 최고 검량농도의 면적을 초과하여 시료를 희석한 후 재분석하였다.


결과 및 고찰
시험기간 중 시설 내 온·습도 및 채취시료의 적합성

녹색꽃양배추 재배기간 동안 시설내의 일평균기온은 18 ± 4°C이었고, 일평균습도는 68 ± 12%이었다. 양상추의 경우는 일평균기온 17 ± 4°C이었고, 일평균습도 66 ± 11%이었다. 상추의 경우는 일평균기온 25.9 ± 6.6°C이었고, 일평균습도 77.0 ± 21.6% 범위였다(Table 1).

수확 작물의 개체별 평균무게는 녹색꽃양배추 217-232 g, 양상추 554-592 g 및 상추 120 g이었고 상품성이 있는 상태로 시험목적에 적합하다고 판단되었다.

분석기기의 재현성, 회수율

토양과 작물 분석법의 기기분석조건에서 검량선은 상관계수 0.99 이상의 양호한 직선성을 보였다.

토양과 작물 중 농약의 회수율은 Table 6에 제시한 바와 같이 74.5-115.1%로 유효한 결과를 보여 확립된 분석법을이용하여 잔류농약을 분석하는데 문제가 없을 것으로 판단되었다.

수확시의 토양과 작물 중 농약잔류량

수확시의 토양과 작물 중 농약잔류량을 Table 7에 나타내었다. 농약의 이론적 처리량은 토양 가비중을 1로 가정하고단위면적당 살포량을 토양 채취 깊이로 나누어 계산하였다. Flutolanil을 예로 살펴보면 기준량 처리의 경우는 6.3 kg a.i./ha인데 시료 채취 토심이 10 cm이므로

6.3 kg a.i. /(10,000 m2)/(0.1 m)/(1,000 kg/m3) = 6.3 mg/kg이다.

토양 중 농약잔류량

Cadusafos, fluopyram, flutolanil, hexaconazole, pencycuron 및 thifluzamide의 농약처리 52-68일(작물 정식 51-65일) 후 작물 수확일의 토양 잔류량은 0.17-3.58 mg/kg이었다. 처리량 대비 잔류율은 pencycuron이 88-93% 수준으로 가장 높았고, cadusafos, fluopyram, thifluzamide, hexaconazole, flutolanil 순으로 감소하였다. 잔류율이 가장 낮은 flutolanil은 초기량의 28-30% 수준인 것으로 나타났으나 단위면적당 처리량이 6.3-12.6 kg a.i./ha로 다른 농약에 비하여 월등히 높아 수확기에도 토양 잔류량은 기준량 처리 1.89 mg/kg과 배량 처리 3.58 mg/kg으로 높게 나타났다.

유럽연합과 일본의 농약평가서에 의하면 시험농약별 야외조건 50% 소실 소요일수(DT50)의 중위수나 평균은 fluopyram 163일, flutolanil 149일, hexaconazole 114일, cadusafos 42일, pencycuron 40일, thifluzamide 39일 순으로 나타나 본시험의 결과와는 부합하는 정도가 낮은 것으로 보인다. 그러나 4-10곳의 장소에서 얻어진 DT50값의 범위가 cadusafos 1-61일, fluopyram 21-539일, pencycuron 11-68일 및 thifluzamide 21-82일이었음을 고려하면 시험결과의 대부분은 정상범위 안에 있다고 생각된다(Appendix 1).

녹색꽃양배추 중 농약잔류량

녹색꽃양배추의 cadusafos 잔류량은 정량한계(0.01 mg/kg) 미만이었는데, 토양에 cadusafos를 9 kg a.i./ha 수준으로 처리한 대사시험 결과 순무잎의 잔류량은 0.018mg/kg 수준(Appendix 2)이었으므로 잔류시험 처리량의 차이를 고려하면 유사한 결과라고 생각된다.

녹색꽃양배추의 fluopyram 잔류량은 기준량 처리구에서 0.01 mg/kg, 배량 처리구에서 0.02mg/kg이었는데, fluopyram을 0.53 kg a.i./ha 수준으로 처리한 동위원소 사용 후작물 잔류시험 결과 근대와 순무잎의 1-3기작의 잔류수준은 각각 0.06-0.30 mg/kg 및 0.06-0.61 mg/kg이었다(Appendix 3). 비슷한 농도로 시험했음에도 잔류수준의 차이가 매우 큰 것은 작물이 흡수·이용 가능한 fluopyram의 양적 차이에 의한 결과일 것으로 생각된다. 작물 근권에 고르게 처리한 근대와 순무 후작물 시험에 비하여 입제를 토양 표면에 처리한 녹색꽃양배추 뿌리 주변 토심의 fluopyram 농도는 매우 낮았을 것인데 낮은 수용해도로 인하여 작물 수확시기까지도 이런 상황은 지속되었을 가능성이 있다(Appendix 1).

양상추 중 농약잔류량

양상추의 flutolanil 잔류량은 기준량 처리구에서 0.01mg/kg, 배량 처리구에서 0.02mg/kg이었는데, flutolanil을2.7 kg a.i./ha 수준으로 처리한 동위원소 사용 후작물 잔류시험 결과 상추와 순무잎의 잔류량은 0.04-0.05 mg/kg 수준이었다(Appendix 4). 양상추 시험의 처리수준이 참고시험의 처리량에 비하여 2.3-4.7배 높음에도 불구하고 잔류량이 후작물 시험에 비하여 현저하게 낮았다. Fluopyram의 경우와 마찬가지로 토양 표면에 살포된 입제 중의 flutolanil이 작물의 근권부로 충분히 이동하지 못한 결과 양상추의 잔류량이 낮게 나타난 것으로 생각된다. Fluopyram 녹색꽃양배추 시험결과와 함께 살펴보면, 작물지상부로 이행의 지표가 되는 옥타놀-물 분배계수(log Pow)가 3.17-3.3으로 매우 유사하고, flutolanil의 살포량이 fluopyram의 10배를 넘는데도 불구하고 두 시험의 작물잔류량은 비슷한 수준이었다. 이것은fluopyram에 비하여 flutolanil의 물에 대한 용해도가 50%, 토양흡착계수는 2-3배 수준이므로 근권에서 작물에 흡수될 수 있는 농도는 비슷한 수준이었을 것으로 생각된다(Appendix 1). 따라서 농약의 토심별 분포 양상에 의해 잔류량의 차이가 크게 영향을 받을 가능성이 있는 fluopyram과 flutolanil은 토양표면처리에 처리한 입제시험 결과로 후작물에 대한 잔류수준을 판단할 수 없다.

양상추의 pencycuron 잔류량은 정량한계(0.01 mg/kg) 미만이었고, pencycuron을 2.5 kg a.i./ha 처리한 동위원소 사용 후작물 잔류시험에서 pencycuron은 1기작 근대에서 0.025 mg/kg 수준으로 잔류된 것을 제외하면 3기작까지 근대와 순무잎에서 0.001-0.006 mg/kg 수준으로 검출되었고(Appendix 5), pencycuron 제품농약을 0.7-2.5 kg a.i./ha 수준으로 처리하고 30일 후에 식재한 양상추, 순무잎 중 pencycuron의 잔류량은 정량한계 (0.01 mg/kg) 미만이었다(Appendix 6). 시험장소가 다른 3곳에서 수행한 포장 후작물시험 5건의 결과가 모두 정량한계(0.01 mg/kg)인 것을 고려하면 pencycuron이 후작물 엽채류에서 0.01 mg/kg을 초과할 가능성은 낮을 것으로 생각된다. 이러한 결과는pencycuron의 물에 대한 용해도가 0.3mg/L로 상당히 낮고, 토양흡착계수(Kfoc)가 2,414-10.441 L/kg 수준으로 매우 강한 특성과도 부합된다(Appendix 1).

상추 중 농약잔류량

상추의 hexaconazole 잔류량은 정식기 처리구에서 정량한계(0.01 mg/kg) 미만, 정식 31일 전 처리구에서 0.02 mg/kg이었다. 일반적인 예상과 다르게 농약처리 기간이 29일이나 앞섰던 처리에서 상추 중 잔류량이 높은 것은 정식 전 30일동안의 수분 공급과 정식 전 경운에 의하여 토양 내 hexaconazole의 분포가 정식기 처리의 경우보다 상추로 흡수되기 용이하였기 때문일 것으로 생각할 수 있다. 그러나 합제로 처리된 thifluzamide의 상추 중 잔류량이 정량한계(0.01 mg/kg) 미만이었고, thifluzamide의 처리량은 hexaconazole 처리량의 절반 수준에 불과하여 thifluzamide의 결과가 hexaconazole의 잔류양상에 대한 추정을 뒷받침하지는 못하였다.


결 론

Cadusafos와 pencycuron은 입제 처리 잔류시험 결과에 의해 후작물 잔류시험의 필요성이 낮은 것으로 판단되었으나, fluopyram과 flutolanil의 경우는 후작물 잔류시험이 필요할 것으로 판단되었다. Hexaconazole과 thifluzamide의 경우는 PBI 30일 처리에서 상추 중 hexaconazole이 0.01 mg/kg을 초과하였으므로 후작물 잔류허용기준을 설정해야 될 것으로 생각된다.
