Abstract

Although non-animal testing has increased internationally, their application in evaluating pesticide formulations remains limited in Korea. Skin and eye irritation of pesticide products are classified, under the Korean pesticide control act, into four categories: severe, moderate, mild, and no irritation. Internationally accepted non-animal test methods provide information that could support classification under the Globally Harmonized System of Classification and Labeling of Chemicals (GHS), under which irritation is classified into Category 1, Category 2, and No Category. Our study compared classification of irritation under the Korean regulation with those obtained by applying the GHS criteria to the same animal study data. For skin irritation, the Korean classification of severe, moderate, and no irritation corresponded to GHS Category 1, Category 2, and No Category, respectively. Mild irritation (2.9%) was classified as GHS No Category. For eye irritation, the classification patterns were similar for the two systems. There were differences for some moderate (2.2%) and mild (2.9%) irritation, as they differed when linked with the skin irritation classification and the criteria for reversibility. Our findings indicate that when existing animal study data are used, the classification of some pesticide formulations might change when transitioning to a GHS-based system. Therefore, potential classification changes must be evaluated prior to regulatory transition. Our study provides a foundation to improve the irritation classification system for pesticide formulations in Korea.

(European Parliment and council, 2008; RDA, 2026a; US EPA, 2026).

(Globally Harmonized System of Classification and Labeling of Chemicals, GHS)은 국가와 산업 분야별로 상이한 화학물질의 유해성 분류 및 정보전달 체계를 국제적으로 조화시키기 위해 마련되었다(UN, 2025).

(Table 1, Table 2).

Classification criteria for skin and eye irritation of agrochemical formulations under the Korean Pesticide Control Act

GHS classification criteria for skin corrosion/irritation and serious eye damage/eye irritation

(Table 1, Table 2).

Comparison of skin and eye irritation classification of pesticide formulations according to the Korean pesticide regulation and GHS criteria (2024-2025)

Simetryn EC, Bentazone-sodium.Fenoxaprop-P-ethyl ME, Pyroxasulfone.

Comparison of Korean and GHS classification of pesticide formulations with skin or eye irritation from 2018-2023

Distribution of irritation classifications for pesticide formulations according to the Korean and GHS criteria (2024−2025)

Additional analysis of skin irritation classification using an independent registration dataset (2018-2023)

Simetryn EC, Cinmethylin.Penoxsulam DC, Fluroxypyr-meptyl. MCPA ME)은 Category 2로 구분되었다(Table 3, Table 5).

Classification criteria for skin and eye irritation of agrochemical formulations under the Korean Pesticide Control Act

Classification Classification criteria under the Korean pesticide control act
Skin irritation Eye irritation
Mean scores are calculated separately for each animal using observations recorded at 24, 48, and 72 hours after application. For delayed skin reactions, observations recorded on three consecutive days after the onset of the reaction are used.
Severe A pesticide with a pH ≤ 2 or ≥ 11.5; or
a pesticide causing skin damage including corrosion or tissue destruction in at least one animal immediately or during the 4-h exposure period, with the effects persisting through Day 14
A skin-corrosive pesticide; or
a pesticide with a pH ≤ 2 or ≥ 11.5; or
a pesticide with effects on the cornea, iris, or conjunctiva in at least one animal that are not expected to reverse or do not fully reverse within 21 days; or
a pesticide with a mean corneal opacity score ≥ 3 or iritis score ≥ 1.5 in at least two of three animals
Moderate A pesticide with a mean erythema/eschar or edema score of ≥ 2.3 and ≤4.0 in at least two of three animals; or
a pesticide causing persistent inflammation including alopecia, hyperkeratosis, hyperplasia, or scaling in at least two animals through the end of the observation period, normally 14 days
A skin-moderate irritant pesticide; or
a pesticide with a mean corneal opacity or iritis score ≥ 1, or conjunctival redness or chemosis score ≥ 2, in at least two of three animals with the effects fully reversing within 21 days
Mild A pesticide with a mean erythema/eschar or edema score of ≥ 1.5 and < 2.3 in at least two of three animals a pesticide with effects meeting the criteria for moderate eye irritation but fully reversing within 7 days
Non-irritant Does not meet the criteria for mild, moderate or severe skin irritation Does not meet the criteria for mild, moderate or severe eye irritation
